Darolutamide Side Effect?

I’m currently taking Nubeqa (daralutamide) without ADT. I’ve also been taking gabapentin for a spinal problem and associated radiculopathy. I’m noticing what I think is muscle wasting and loss of strength in my legs, and although I think it’s more likely related to the spinal problem &/or gabapentin, I was wondering if anyone has had a similar experience with darolutamide or gabapentin?

I would expect gynecomastia as a side effect if you do have no ADT. Take tamoxifen 20 mg against that.

I’ve been on darolutamide for 3 months and have mild gynecomastia and mild breast tenderness. Good to know there’s a remedy if it worsens, but probably not worthy of taking another medication yet with it’s own set of side effects.

I took two 3-month courses of a similar drug, enzalutamide (Xtandi) as part of a clinical trial at NIH. It stopped working for me and my MO had me start darolutamide in November. My back issue began in December and I’ve dealt with a lot of pain and have been taking gabapentin for the last 2 months. The only SE I’ve noticed definitely attributable to darolutamide would be the mild enlargement and tenderness below the nipple, which apparently responds to tamoxifen if needed.

Great thank you for sharing your experience. I too was on Xtandi but could not tolerate the side effects. Hoping for a better experience with the darolutamide. I had radiation to my breast tissue last year so that should not be a problem for me.

Hi GP, so Lupron reduces the chance of gynecomastia when taking Darolutamide?

Darolutamide does not reduce testosterone and estrogen, even increases the level of these. Estrogen causes the breast to grow and Tamoxifen blocks this.

If you add Lupron, it reduces both testosterone and estrogen and there is much less estrogen available to let the breast grow. Simply put.

At the recommendation of people on this site, and approval of my MO, I’m now on tamoxifen. 3 weeks now, no change. Starting to look ‘hot’ in sweaters. 😢. Will gynecomastia reverse or just slow down?

20 mg of Tamoxifen may reduce the gynecomastia a bit if the tissue has just grown. Otherwise it will stop the breast from growing further.

I was taking gadapentin for a couple of weeks for my neuropathy and had to stop it due to chronic constipation and loss of bladder control. Once I stopped the gadapentin those side effects stopped. I believe it made my lower extremity and pelvic musculature weaken.

Hi Allen, I have been on Darolutamide for about 6 months. The main SE I notice is fatigue for a couple of hours after taking it. I don’t think I have gynecomastia yet, don’t notice and changes or tenderness). I also had some weight gain. I was on Lupron only (I had to stop taking Abiraterone because of high liver enzymes) and I put on 10 pounds and when I added Darolutamide I added another 5 pounds. That’s about it. I think Daro has some similar SEs as Lupron so I only noticed the fatigue and weight gain separately from Lupron.

Gabapentin--been taking for maybe twenty years. Just now quitting--many sideaffects are listed such as frailty, balance, dizzy--all of which I have, so stopping may help.

I took darolutamide for one month. I switched from apalutamide which gave me a lot of nausea and a little depression.

The darolutamide increased my depression. I also found myself urinating in my bed once a week. So, I switched back to apalutamide.
