One week ago Sept 23 I started the Talapro 2 clinical trial (talazoprib/placebo with enzalutamide) and immediately jetted of to Hawaii. Notice a bit more fatigue than just the lupron. And maybe a bit looser bowl. But so far all side effects are very manageable. PSA was at 14, mets to pelvic bone, L4, ribs, femur and skull. Next blood test on Oct 8. I'll keep you posted on how things are going.

Does your cancer has BRCA or ATM mutations? Talazoparib is a PARP inhibitor. Olaparib another PARP inhibitor has shown efficacy in cancers with BRCA , ATM and other DNA repair mutations.

Freedom one said it was inconclusive but all indicators is BRCA mutation likely.

my dad carries the ATM defect and tried a talazoparib + avelumab trial early this year. it was a failure for all ATM patients, they were kicked off. BRCA patients were responding though, so best of luck!

Most cancers with BRCA respond to olaparib (77% had a decrease in PSA) and most cancers with ATM do not respond (only 5.3% had a PSA decrease).

I’m on the talazoparib+Avelumab trial since Late April. BRCA2.

So far scans show the disease progression to the bones has been halted. ALP down to 90 but PSA holding around17-20. (?)

Worst side was anemia

i’m so glad it worked for part of the trial participants!! late april is when my dad got booted from the same trial. onco said the BRCA patients were doing much better than ATM. dad felt almost immediately better after ditching the talazoparib. had a pretty rough infusion reaction the first time, too. fine the second. there was no third.
