So it seems I’ll be starting a trial soon. I’m now off Enzalutamide to get it out of my system. I’ll be switching to Zytiga and getting treated with CPI-1205 (ProStar) in addition through a trial. If anyone is on this regimen or has been in the past it would be great to get some tips. Thanks.

While not yet confirmed it appears I will also be entering the Prostar trial. I have been on Zytiga/Prednisone for 16 months and it has quit working. I will be in the Enzalutamide arm of the trial. I am thankful it is not a randomized trial with placebo at least we will be getting actual medication that the standard of care arena.

It is a randomized trial: "Patients in phase 2 will receive CPI-1205 at the RP2D in combination with either enzalutamide or abiraterone/prednisone vs either enzalutamide or abiraterone/prednisone as a control arm."

Right I understood that...in my case I have been on abiraterone/prednisone so will go to enzalutamide with or without CPI-1205. What I intended to say was we would get real medication and not be randomized to a placebo that would not help me with rising PSA/mets.

Gotcha. They have to give standard of care in the control arm.

I went off the trial a week ago today. I was on Enzo & CPI-1205 since October and my PSA went up every check. Bone scans are fairly stable but a few mets show more intense. The side effects were bad...I feel so much better after a week off. I start the 1st Cycle of Chemo on March 4th...the next standard of care.

Are you on the ProStar trial?

Just signed up for it. I have been on zytiga since diagnosis in August of 2018.

Do you know if you had the active drug or placebo?

I was on Zytiga for 1 1/2 years with no real side effects but when it failed we looked at the trial. I first went to Enzo (and had side effects) with no positive results. That lasted about 3 months we then restarted with Enzo & CPI 1205 (there is no placebo in this trial). After 4 months of no positive effect and a lot of side effects we quit the trial. I wish you well with it I may have been an anomaly. If your at Duke then you are working with Christine Daly...she was great to work with.

I go to UF in Jacksonville. It’s in stage 2 now, so patients get randomized to Xtandi + CPI-1205 or Xtandi + placebo. So I won’t know. Not sure how I feel about that

Ok...I thought the stage 2 was randomized to Xtandi or Xtandi + CPI 1205 (actually for those that were on Xtandi it would be Zytiga or Zytiga + CPI 1205.

I see Dr Dan George at Duke. I know I was randomized to the Xtandi arm then did a cross over to Xtandi + CPI 1205 when Xtandi did not work on it's own. I have been told there is no placebo involved.
