Levodopa and CYP3A4, but probably not Grapefruit juice... The next steps
Partially inhibiting CYP3A4 may help patients lead a bettter life with less "off times". 2) The on/off effect may have long-term implications for
dyskinesia
, posssibly causing damage to synapses that must adapt to repeated under/over dosing of dopamine.

Why should not we increase L-Dopa bioavailability in the Brain
After watching this video you will find out why you shouldn’t manipulate the L Dopa bioavailability in the brain. https://youtu.be/_eq28PCvfsg You are trying to balance something but you will end up messing with the same thing, in this case the Dopamine.

Artificial sweetened Cola drinks rapid relief?
Hi, i was dx with PD in 2014, and started out with selegilin and Requip (dopamin agonist), later adding L-DOPA (Madopar/Sinemet) and entakapon. Altogether enabling me to work full time, and I am convinced this would not be possible without such a «cocktail». Still, episodes of fatigue with loss

Sinemet suddenly not working well
Any advice would be welcome. I take 3 100/25 sinemet a day an have been since January. Before that I was taking 2 a day for 6 months. I also take rasagiline 1mg. The sinemet was making a real difference for me for 4 hours at a time 8am. 12 and 4pm until the last 3 weeks where now it only lasts 1 to 2

July 24, 2019. NOT “ESSENTIALLY SYMPTOM FREE”
Dyskinesia
is embarrassing. As with so many people with PD, my body stiffens painfully if I stay too long in one position. I couldn’t bear the thought of flying to Kyoto. I could go on, but I would rather focus on positives.

Wallowing in the Trough of Carbidopa and Benserazide
I hesitate to even start a thread that is heresy to many, many PD sufferers, many self proclaimed pharmacologists/biochemists on this forum and many of their physicians as well. It is all about a couple of PD drugs that can go very deadly if used inappropriately. Please realize, if you can do so, that

Sinemet, how much is too much?
I feel like I'm on the border of my limits as more than 3 1/2 pills a day cause me some
dyskinesia
(head movement notable) and that's annoying and do not allow me to sleep.

Does anyone have any sleeping remedies or meds that work?
I was taking Endep (which gave me a full night's sleep) but my Neurologist changed me from Stalevo (which was giving me Dyskenesia) to Madopar + Xadago. But I had to cease the Endep as he said it could conflict with the Xadago, which is a fairly new drug that is still being trialled. He prescribed Paxam

More pieces to the puzzle about getting a more stable supply of levodopa to the brain
Here is my latest article about how the CYP3A4 enzyme metabolises levodopa and controls its bio-availability and haf-life. It is based on a 50-day cycle (n=1) during which CYP3A4 was alternatively non-inhibited and inhibited. The results confirm that CYP3A4 powerfully inhibits levodopa and may be more

Age old question: does PD or L/C cause Dyskinesia?
I know there are recent studies out there that say that L/C doesn't - that PD's progression does. So someone just diagnosed might as well start on L/C straight away, right? But does anyone really know? Are the studies funded by the drug companies somehow? Asking for a recently diagnosed friend

Doctors use levodopa responsiveness to diagnose, but a new report today says a specific gut bacteria prevents it from getting to the brain
I often feel I am sometimes unresponsive to levodopa. I wonder if these bacteria come and go. In addition to the most important matter of controlling symptoms, another place this matters is that we could be excluded from clinical trials for being unresponsive to levodopa when it is not because we do

Comparison of selegiline and levodopa combination therapy versus levodopa monotherapy in the treatment of Parkinson's disease: a meta-analys
S + L combination therapy is superior to L monotherapy for the improvement of clinical symptoms in PD patients. Moreover, the safety profile of S + L combination therapy is comparable with that of L monotherapy. https://www.ncbi.nlm.nih.gov/pubmed/31175606

Something interesting
Today I had a colonoscopy. Friday I stopped all the PD medication including the vitamins and I ate a little and after that nothing for a day and I cleansed my digestive system. Guess what: all my symptoms almost gone, just a little tremor left on my RH. The tremor intensified just before the procedure
