The FDA has approved itovebi (inavolisib) plus Ibrance (palbociclib) and Faslodex (fulvestrant) ...is anyone taking this trio?

If you are taking it, had you previously used Ibrance until it failed? I'm on Piqray and faslodex right now but feeling like the end of this run is near so Im wondering g if there is anyone who has successfully returned to Ibrance with the addition of itovebi (inavolisib) and faslodex? Such a grab bag of drugs available now but not all are working if you have the PKC3A mutation...wishing you a the very best!

Looks really good in terms of efficacy. Interesting that adding it to a SERD and targeted therapy overcomes endocrine resistance. Also good that it is oral, not infusion. Only for those with the PIK3 mutation.

Under the current guidelines, though, I don't think you would qualify. It was not approved for those who have had prior systemic therapy.

thank you…so at this time they are still looking to see how well itovebi works on a “virgin” cancer …? Sounds so heartbreaking to be put on that shelf of failures. Well, fingers crossed that Piqray continues to work. Thank you again…❤️‍🩹❤️‍🩹

No, as I understand it, it has been approved for people with bc who have been on a medication to reduce estrogen (and that alone) and it didn't work. It does not seem to have been approved for those who have been on "systemic" treatment. Perhaps that is what the research explored and it got fast-tracked for use with that population. Maybe that is the population it was tested on or maybe that is the group it worked for. If you were to get it, I guess it would be off label.

But look, Cazzandraus was on Ibrance first, and that is targeted....is that the same as systemic?

Thank you again Tammy…I did a little checking on the terminology of systemic…both Piqray and IBrance are considered targeted.. whew…when I was considering the new med after IBrance, I first thought Xeloda was a safer choice than Piqray…but Xeloda is not targeted. It’s an oral chemo that works systemically. That being said, my ONC suggested that returning to IBrance is not as successful as it was on the first go. However, if itovebi can work with fulvestrant ,… well, fingers crossed. We are entering new territory in this fight. Thank you again!

So good, you are eligible. Lucky you. I am on Xeloda, so I am out. I also do not have the PIK3 mutation. (My onc would not let me return to Ibrance, despite what Dana Farber found about taking a break and trying again. Instead, I went on the alternatives, Verzenio and Kisqali. They did little or nothing.)

I have been taking this medication with just Fulvestrant for almost 4 years now. I was in one of the clinical trials. I was on Ibrance first and then moved to this drug. It has way less side effects than Piqray. I would still reach out to your doctor and discuss this option, it can’t hurt!
