The Take Home Message on Cujoe's Posting on CRISPR edited T cells--THIS IS HUGE AND IMPORTANT

This was the first EVER, sanctioned by the government, multiple gene editings to the human genome by a company and the results were:

First, we can successfully perform multiple edits with precision during manufacturing, with the resulting cells surviving longer in the human body than any previously published data have shown. Second, thus far, these cells have shown a sustained ability to attack and kill tumors.

One of the editings was the removal of the checkpoint, PD-1...Checkpoints are what cause the slowing down of immune attack because cancers--demons that they are--can disguise themselves like wolves in sheep's clothing--see easy explanation below:

By doing this, they can infuse T cells that will provide a sustained attack and will not be so easily tricked by cancer cells not to continue the attack...Secondly, there are drugs in development to stimulate macrophage attack against cancer cells which I have posted on--see below:

Could a drug like CpG be added to these engineered T cells to further the T cell attack by getting macrophages involved? Absolutely, IMHO--a double attack...

Finally, could an infusion of these engineered T cells be combined with CpG or a CLEVER-1 inhibitor AND...pick one:

1) Checkpoint inhibitor

2) PARP inhibitor

3) Lu-177 or AC-225

4) SM-88, a metabolic drug under development...

5) There are others under development...or here--chemo, etc..

Thus, a 3 pronged attack...or more...

Yes Don. It is definitely COMING. CRISPR is the future of medicine. About time those overpaid pompous quacks got technical.... Real science at last.

Yes, there were few comments to cujoe posting the information...the science is a bit overwhelming for some, so I hoped to provide some illumination to what the CRISPR edited T cells offered in potential...

Despite all of this science, Marnie had a posting where Maha Hussain, MD of Northwestern said that 70% of Urologists started people on Lupron ONLY as first line treatment and that is wrong as proven by STAMPEDE, LATTITUDE, etc... and we wonder why the ACS lists the 5 year OS for Stage4 as 30%... what would a simple change like using Zytiga first with Lupron make to OS??

The Science is great, but without changes to treatment protocols by local Urologists, then we are screwed... It is why I say to newbies, always....ALWAYS... get someone that specializes in PCa... it can save your life IMHO...

Keep things running in the UK, my man...

For a great run down on CRISPR, try to log in to BBC iPlayer (on the BBC website). You will need a VPN outside the UK to do this. Then look for STORYVILLE - THE GENE REVOLUTION...... This is an excellent presentation of the technology and it's potential...

Like CAR-T, there are no approvals for solid tumors, but with this CRISPR technology, will we see 6th Generation CAR-T with PD-1 or PD-L1 checkpoint removed and a PSMA ligand?? IMHO, I believe so...the developments in immunotherapy continue and I believe we shall soon see some solid tumor testing in Prostate cancer... The hunt continues...
