Another possible breakthough currently in phase 1 human trials ? - or false dawn ?


Here is the 2018 pubmed link on its mechanism of action. Seems very promising for all sorts of solid cancers.
German cancer researchers are rather skeptical about the chances of success of the AOH1996 study. Preclinical data are usually not suitable for predicting success in clinical application. The differences between tumor growth in cell cultures or mouse models on the one hand and the growth conditions of a tumor in the patient are too great.
One of the German cancer researchers points out that the antitumor activity in the mouse models used was rather moderate, and in one model it was downright weak. In addition, the mouse tumor models used would be considered particularly poorly informative for clinical reality, so they would only be a first approximation.
On closer inspection, 70 would turn out to be only nine types of cancer. The substance was tested in 70 cell lines representing nine different types of cancer. Within each cancer type, a very heterogeneous reaction to the treatment was found. Translating this into a prediction of clinical efficacy means that not all cancers or patients will respond well. The type of cancer that can be treated particularly well has not yet been found, they say.
The German cancer researchers therefore think that AOH1996 actually needs several more years of research and development, and that the early start of a phase I clinical trial with only 8 participants is likely to cause the trial to fail early.
