I heard back from City of Hope that I have strong PSCA expression, and I'm moving forward with the CAR-T trial. I thought I'd share this video clip from the office to celebrate.

Brilliant news. I hope all goes very, very well. Just a quick question if ok with you - how do they test for PSCA expression?

Immunohistochemical (IHC) staining of the original biopsy sample. Of course that leaves the question of whether the expression level is the same in the newer metastatic growth.

My understanding is that the killing of PSCA expressing cells releases other antigens that are presented to the immune system to flag non-PSCA presenting cancerous cells as foreign. Time will tell how effective the crossover is.

I've said it before I appreciate your contributions here and your diligence leading to further understanding our PC and treatments. I am sorry that yours is a fast moving Pc but again thank you for doing that which may be an option down the road for myself and others.Im counting on good results for you!

Best of luck Jav,I re-read your Bio. PSA never above 2.5 !

You have tried everything. Lu-177, Cabazataxel, Docetaxel. PCa should tap out soon and say….enough.

Please keep us posted on progress. Many of the smarter guys on HU think Car-T may evolve to create remission.

Been through docetaxel, lu -177, zytiga, cabaxitaxel and now starting xtandi. Still on Lupron. Hopefully CAR-T does the trick.

I am so happy for you getting into this trial. My brother was three days from getting Leukophorisis for a Car T trial at Dana Farber. I am following your journey closely as I truly believe you will get good results. Thank you so much for taking part in this trial and giving so many hope!

I don't think it's ductal. It was metastatic to the bone, and castration resistant at diagnosis. So far no organ or lymph node involvement. That gives me hope.

I wonder if you know, how common is “high enough” PSCA expression to qualify for this particular trial? i.e. do you have any insight wrt how many candidates were disqualified due to inadequate PSCA?

Thanks. I don't know what the cutoff is for PSCA expression. 80 % of my biopsy sample expressed PSCA. They called that very good.

Following your experience with all fingers crossed and all prayers said. My husband has CAR-T on his radar for CLL. There have been good results with blood cancers. God speed!

Jav, what is your schedule at City of Hope, and when do they look for results. With SABR/SBRT, it is at 3 or 4 mos with results 12-18 mos seeing improvement. What do they hope for with Car-T?Thanks,

My records are being submitted to the sponsor this week for approval. They're targeting May 18 for harvesting my T cells (leukopheresis). It takes about 4 weeks to grow the cells and then infuse them.

I'm scheduled to receive two doses if the first doesn't wipe it out. Second dose would be around 6 weeks later.

I think the effects are pretty quick, since the Cytokine Release Syndrome (CRS) lasts a few days. I need to stay close to City of Hope for a month after infusion.
