Advice /Experience with AMG 160

Hello,

Thank you for all the information this group shares! You will see from my profile I have been through all the SOC drugs and LU177 and AC 225 and am now looking for next treatment option. Looking for advice/experience with a trial with AMG 160, does anyone have any feedback on their experience? Or do you think chemo would be better option (did okay with docetaxel side effects but it did not resist cancer growth for too long). Thank you, Lowbed

I'm sort of surprised anyone is recommending a PSMA-targeted BiTE therapy to you if Lu177PSMA and Ac225PSMA failed. In fact, that is listed as an exclusion from the AMG160 trial.

Any hints from biopsy of a metastasis?

Cabazitaxel+carboplatin may be worth a discussion with your MO.

TA-thanks very much for your viewpoint which I greatly value. LU 177 worked for 14 months and AC 225 J591 worked for 6 months. I was thinking that if I got 6 months from AM 160 that would string time along. I got an exception to the exclusion. I have had several biopsy and liquid biopsy ( blood, lymph node tissue and blood)reviewed by Foundation One and nothing has come back to work as a match with any available drugs. Thank you

If your PSMA therapies stopped working, why would it still be PSMA-vulnerable? Have you had an FDG PET? What about IHC analysis of tumor tissue? What proteins is it expressing or not?

Thank you for your message, we meet with the Dr. today and will ask about the IHC analysis. We have not had a FDG PET. I thought the PSMA therapies stopped working because you are not getting the drug anymore because of limits on radiation dosage. Thank you

Were you told they reached a radiation limit? Did salivary side effects become troublesome?

Hi, thank you for your responses, your knowledge is incredible. At the oncology appointment today, we were surprised to learn that a wait list spot for an androgen degrader trial came open today- if eligible we will go on that trial. We were not specifically told that we hit radiation limit. Two infusions with AC 225 591 in February did have salivary side effects that have mostly gone away except for dry month and odd metallic test sometimes. Thank you

Would that new trial be for a SARM? A Selective Androgen Receptor Modulator? Very interesting newer approach recently discussed on this forum. Good luck.

My guess is that it is one of the BET Bromodomain inhibitor trials, like ZEN-3694 or several others. It's a very promising area of investigation.

The drug is called CC-94676 by Celgene.

My husband did the AMG 160 trail it did not work for him. He got through 4 rounds and it was really hard for him. After that they did a scan because he was so weak and in a lot of pain. It did not work he’s cancer has spread to his liver and brain. He is in home hospice now.

Very sorry to hear that your dear husband's gallant fight against the beast has gone awry. I must tell you that my mother used to say "feed the birds and you'll never go hungry". God Bless you and God Bless your husband.
