Hi guys just looking for some reassurance my mum started the niraparib 2 weeks ago the oncologist not her usual one wasn't going to let her start it as her CA125 had risen but a 2nd opinion advised her to start it. So at the start of the diagnosis her CA125 was 40000 she got it down to 141 with chemos and op before starting the tablets it had creeped up to 165 and again has went up again only slightly they did say its early days but my mum is devastated hopefully it's just a wee blip and it will level off soon.

I think that you and your Mum will probably have to wait and see unfortunately. Ca125 rises can be for all number of reasons, including, (I believe I’ve seen this somewhere) the action of Parp inhibitors when you are first taking them. But…and it’s a big but, clearly you can’t ignore the possibility that her cancer is active again. Will she get a CT scan at some point? This might be the best way forward..at least she will get some idea of what is happening. I always think the not knowing is the worst xx
You say she's only been on Niraparib for two weeks? If that's correct, that is not long enough for it to do anything much - it can take 4-6 weeks to see whether its starting to be effective or not. When I went on it, it was about 6 weeks till it looked like it was plateauing, and 8 weeks till we saw a small drop in the CA125, so it's not time to worry too much yet...

Mine continued to rise for a couple of months after I started taking it but then started to very slowly drop. However, I was not put on it until 12 weeks after chemo when it should have been within 8 weeks so they attributed the delayed response to that. Two weeks is a very short time. Stick with it and keep your hopes high. Any sort of infection and even the covid jab can make the CA125 rise temporarily x
Hiya thanks for getting back that's reassuring as again my mum was 10 weeks when it should've been 8 but due to low magnesium she was so tired they gave her an extra 2 weeks to recover so hopefully that's what it's been .
