just got my annual review results back MPL Et here. Pre Pegysus it was 28 % 2 years ago. Last year 30% this year 32%. Last year haematologist wasn’t concerned. Said could just be day of the week kind of change. Will ask relevance when I see them in 6 weeks but is it significant? Was hoping the allele burden would reduce with peg. Any ideas. My last blood count was wildly out butI was I’ll. respiratory infection when it was taken.

allele burden VAF rising
There is minimal data on IFN with MPL. This study with Besremi will be looking at it:

"Quantitative measurements of JAK2, CALR, and MPL allelic burden will be undertaken by a central laboratory every 6 months"
This another study including MPL with different details:

There is this study that had one pt completely eliminate MPL with Rux. Of course this single outcome does not drive decisions but is interesting:

In the meantime there is not much in research to go on for mutation level, IFN and MPL. But as your Dr notes, the changes are small. In another post you noted one was via BMB and another was blood draw. Best to compare only with the same method. My Jak2 varied by 5 points between the methods.
