Calr difference type 1 and 2?
I posted today, but deep in a different thread.
Here is some basic info:
It is a complex genetic variation as seen in the report below. One type has something missing, the other has something added. There have been studies of the prognosis for each, the reports are not conclusive or are conflicting from what I have seen. This study found type 2 had higher PLT.
The terms type 1 and 2 are starting to fall out of favor lately as they are finding more CALR variants.

Hey Green...
As EP Guys stated below, it all tends to be a tad convoluted. However, CALR Types 1 & 2, tend to account for over 80% (approx') of CALR mutations... However, there are over 50 different types & counting, as I understand it...
The simple thing to remember is that ALL the so-called, "Driver" mutations, switch on the "JAK-Stat" pathway, to a greater or lesser degree... Those 'Driver' mutations:
* JAK2
* CALR
* MPL
* & Unclassified
However, a person can also be what is termed as, 'Triple Negative' in other words having none of the known 'Drivers' at this stage.
It's also important to remember that "Driver" mutations, beginning w/ JAK2 only became known of since 2005; MPL- 2005-6 & CALR 2013. Hence, the science is still quite young...
I am Post ET / MF, CALR Type2, & through my own independent research thus far:
*Type 1 - Usually has a lower Platelet regime & is more benign that sense
* Type 2 - Usually has a much higher Platelet regime, & one can suffer from TIAs as a result, in my view
Personally, I've already had two or three Transient Ischemic Attacks (TIAs), which might be directly linked to having a Higher Platelet regime.
I also have what is known as, Von Willebrands Syndrome (VWS), some prefer to call it (Disease), which usually means that certain Proteins are missing in one's blood. In my case, they suggest that my VWS is the Type that is 'Acquired' as opposed to genetically inherited, & quite possibly caused by my Higher Platelet regime also(?)
Lots of mights & maybes in there too...
Anyways, hope this helps a tad...
Best wishes
Steve
