Since my husband started synoclein a few days ago, he has been complaining of constipation. Anyone else have that?

I guess you are meaning "Syncolein", which is mannitol with additive aimed at reducing the gaz (alpha-galactosidase) - and make some money with the distress of people with PD.
Mannitol alone never caused constipation, on the contrary.
I don't know what the specific cause is in your case but you probably don't need one sinc constipation comes with PD anyway. Mannitol may be helpful but consuming more than 25 g./day is stated to be a high risk for diarrhoea. Mannitol is a polyol, which we cannot
digest but which some gut bacteria can. When they do they create lots of carbon dioxide gas and make the stools very soft and liquid! You can Take 20 g or less but be prepared for hard choices between PD symptoms and diarrhoea. (See #Xenos below).
Syncolein seemes to help me be less constipation?? I take a bit less than 1 scoop a day first thing in the morning with some lightly caffeinated tea
