Xtandi (enzalutamide) from India
Has anyone ordered
enzalutamide
from India? If so were there any problems, and what pharmacy did you use? There seems to be a plethora of them.

Survival outcomes and prognostic factors for first-line abiraterone acetate or enzalutamide in patients with metastatic CRPC
Patients with mCRPC who received first-line ARATs had better survival if they had a PSA nadir≦2 ng/mL or a TTN≧7 months. Further study is needed to determine if an early switch in therapy for those in whom neither is achieved may impact OS. https://bmccancer.biomedcentral.com/articles/10.1186/s12885

Supraphysiological Androgens [SPA] Promote Tumor Suppressive Activity of the AR Through cMYC Repression and Recruitment of the DREAM Complex
Thus, we tested sequential treatment of prostate cancer models with SPA followed by the AR inhibitor
enzalutamide
. Our results indicate that acquired resistance to SPA can be overcome by treatment with
enzalutamide
, which induced upregulation of AR and increased susceptibility to SPA.”

The CRP:Albumin ratio & CRPC Survival
Dividing the sample according to the first line treatment chosen, we found OS of 26.25 vs 5.9 months (p < 0.05), 27.71 vs 22.57 months (p < 0.05) and 27.36 vs 23.75 months (p = 0.12), for docetaxel, abiraterone and
enzalutamide
, respectively.

ED & zero libido
Instead of celebrating I am concerned that my treatment (Degaralix &
Enzalutamide
) might be loosing its effect. The past couple of oncology reviews have seen my PSA rising though the oncologist isn’t concerned. A bit of a quandary.

Casodex after castration resistance?
Wouldn't it negatively affect the effectiveness of
Enzalutamide
when/if my dad will have to use it in future considering they act using similar mechanisms? 2) Are those fluctuations in the level of testosterone normal for someone being on Zoladex for several years?

Enzalutamide/Xtandi monotherapy as ADT with fewer side effects than Lupron
Enzalutamide
monotherapy could probably be used instead of Lupron after radiation too.

Any experience with fever please?
Treatments have been RPT in 15, 1st line ADT, RT to prostatic bed in 19,
enzalutamide
, 6 cycles Docetaxel ending Jan 23, Olaparib until April, currently on Aberaterone. PSA jumped from 2.8 July 22 to 729 March 23. It seems Chemo and Olaparib weren’t effective.

A new molecule to check: ADA‑308 in vivo proved to be a potent AR inhibitor against enzalutamide‑resistant prostate cancer
Ok, we must wait at least for a phase 1 study, but since the molecule has already a name it makes me think it has been patented already, so clinical studies should not be far away. https://www.spandidos-publications.com/10.3892/or.2024.8791

Enzalutamide at half strength?
I have high volume mrpca and have had triplit therapy with
enzalutamide
at 160 mg per day. My response has been great with PCA falling from 700 at diagnosis to undetectable at 3 measurements over the last 6 months.

Enzalutamide or Darolutamide
Hi, I have advanced metastatic hormone sensitive prostate cancer and as well as my usual three monthly ADT (Lucrin-Depot) injection, I have been offered either
Enzalutamide
(Xtandi) or Darolutamide (Nubeqa). My PSA is currently below 0.1 due to the Lucrin-Depot.

Determinants of PSA Response to Bipolar Androgen Therapy [BAT]
Responders were more likely to have had a radical prostatectomy as definitive therapy and were more likely to have been treated with an androgen receptor (AR) antagonist (
enzalutamide
or apalutamide) immediately prior to BAT (compared to abiraterone).

Triplet Regimen vs Clinical Trial (Pluvicto)
I was given information on apalutamide, darolutamide,
enzalutamide
, and abiraterone acetate. I’m not sure if I will have a choice, but I’d love to know if anyone has experience or opinions, especially if your disease has manifested similarly to mine.

BAT in Argentina
Interestingly, the PSA-PFS for
enzalutamide
increased from 3.8 months after abiraterone to 10.9 months after BAT, and PFS2 with BAT followed by
enzalutamide
was significantly longer than with
enzalutamide
followed by BAT, proving the hypothesis that BAT can re-sensitise CRPC to subsequent antiandrogen
