Motegrity helped me with the off time

I got prescription for Motegrity (2 mg) to help me with the constipation. It is effective and it positively changed how my Parkinson medication works. I could reduce my dose which helped with dyskenisia. It also greatly improved the off time frequency and severely.

From the FDA label:

shirecontent.com/PI/PDFs/MO...

" 12.1Mechanism of Action

Prucalopride [Motegrity] , a selective serotonin type 4 (5-HT4) receptor agonist... In isolated GI tissues from various animal species, prucalopride facilitated acetylcholine release..."

sciencedirect.com/topics/ne...

Serotonin heading:

" 5-HT4 receptor–binding sites are localized with high density in the striatum, substantia nigra, and olfactory tubercle and have been reported in the hippocampus as well .... The 5-HT4 receptor is located postsynaptically to serotonergic neurons and modulates the release of several neurotransmitters (i.e., acetylcholine, dopamine and GABA)," [Emphases added]

You just found a new Parkinson's treatment.

Park_bear, thank you for this interesting information. For roughly 2 weeks after I started on this medication I didn't experienced any significant OFF periods. My sensitivity to animal protein seemed to be reduced. I thought that I could cut down on my Parkinson medication and sure enough for the past 3 weeks now for my two mid-day treatments I take only 1 Rytary pill instead of 2. This significantly helped with the dyskenisia. I noticed however that now I frequently remember my dreams (nightmares) which happened in the past very seldom. I wish I had 100 mg Rytary in addition to 200 mg I am currently on so that I could fine tune C/L intake.

Will your M.D. prescribe it if you request it? S/he would probably be interested to know about how the Motegrity is helping.

I got it from gastroentorologist. I know he communicated with my neurologist. I am going to see neurologist in January and I will tell him. The medication is pricey: I paid $220 for 30. I am not sure if it crossing bbb. There was publication comparing linzess to motegrity in pwp and linzess has shown even better results.

The improvement that you are experiencing is evidence that it is crossing the BBB.
