Very interesting. The link between the JAK-STAT and RAS-MAPK pathways has been known for some time. The RAS-MAPK pathway is downstream from JAK-STAT as illustrated in this article. It appears that the authors have isolated some of the mechanics of how this process works and the impact on apoptosis. According to the authors, this could lead to a new treatment option involving DUSP1 (Dual Specificity Phosphatase 1) expression.

These kinase systems are quite complex. Understanding them requires in depth knowledge of proteomics and more. It will be very interesting to see if this research move forward.

I think that is a relativistic statement. The more full observation would be that JAK2 inhibitors are not thought to cure the underlying disease, but do help to control the symptoms. The hope seems to be that by targeting DUSP1 there is potential for a curative approach. Whether this hope is raliatic is to be determined.

I absolutely agree.

Although, there also seem to be a lot of potential problems possible with the use JAK2 inhibitors, but given the limited choices, sometimes they're the only/best option, or at least the lesser of several evils.

As for me, for now, I'm still betting on Besremi, L. Gluthianone, and taking my B. Alanine in the morning for symptomatic treatment. [While it's not 100%, it does seem to give me some decreased pruritus.,..].
