hi Lynne, sounds to me that your Jak 2 gene is normal, and your Calr gene is the one that’s not working as normal, maybe others can elucidate. Mine is like that too.

There are three known driver mutations that cause MPNs. JAK2 = Janus Kinase 2. CALR = Calreticulin. MPL = Myeloproliferative Leukemia Virus.
JAK2v617f is the most common. There is also a variant - JAK2 Exon 12. The JAK2 mutations cause both PV and ET.
MPL is the least common. It is associated with ET.
CALR is the second most common type of mutation found with ET. It is rarely associated with PV. There are two known types of CALR mutation - Type 1 and Type 2.
Each of these mutations have common in that they deregulate the JAK-STAT pathway, one of the body's kinase systems responsible for hematopoiesis, inflammation, and more. However, these mutations are distinct entities and may have different presentations of the MPN. There is ongoing research into these mutations and the role they play in how MPNs present and should be treated.
