If I miss taking my sinemet it seems there is no noticeable difference in my physical or mental state. Yesterday I forgot to take two sememet and I seemed to perform better this morning. Can anyone suggest what is going on ?
My normal medication is 4 x100+25 sinemet.3x ropinerole,1x azilect,3x pregablin,

I would advise you that you should accept that sometimes you can get relatively better in real terms and try to see if you can work out how to uphold it for longer. That's what I do.

Totally agree. If early to this disease you can go thru a period where it feels like levodopa is practically pointless and a month later it’s absolutely required. Stress, diet, internal dopamine production and other factors change all the time. It’s a bit like throwing a dart at a dartboard attached to a F1 car sometimes.
This is fairly common for PwP. (And indeed very common if you include the pill-wise, overnight fast.) Each time that you take a pill things are slightly different. When these variations stack up in your favour you feel good and you don't feel the need to take all your doses: stress has the effect of burning up levodopa, and v.v; exercise; the workings of your gut; the impact of when you eat with when you take levodopa; the variation in the pills.
Levodopa has a short half-life so it never reaches steady state.
