The essay and PPT fail to mention the locus coeruleus (ceruleus) and the nucleus basalis. The historical focus on the substantia nigra has become outmoded as a ruling model, though the SN is still important, as are treatments for motor symptoms, also the newer model needs include other neurotransmitters, their affected nuclei, and related non-motor symptoms.

Neuronal loss is greater in the locus coeruleus than nucleus basalis and substantia nigra in Alzheimer and Parkinson diseases.
