I take some GABA with every dose of Sinemet that I need. I believe in "listening to my body". I do not adhere to a schedule for PD medication. When a tremor in my arm gets into my pectoral muscles is when I know it is the right time to take a dose of Sinemet. it would be foolish of me to say what is the best level of GABA to seek, I am not a scientist. I have no idea how much of the GABA I ingest makes to the tissue of my brain anyway. But something about taking it managed to help me manage my tremor, I know that is true. I can say the product I buy and how large the capsule is and you can experiment with it yourself. I buy SWANSON"S GABA-plex. I began experimenting with GABA about 7 months ago, found it helped a little. Tried the GABA-plex product, found it helped a little more. Do not buy out SWANSON's inventory and drive up the cost :) I am on a fixed income.

By the way, I am also prescribed Mirapex to help me with the restless legs that used to wake me up at night. Since beginning to supplement Sinemet with GABA-plex the jumpy legs decreased and I successfully quit Mirapex by just taking a half a Benadryl at night along with it and gradually decreasing the amount of Mirapex. It took a couple of weeks to taper off Mirapex. I am glad to report that since quitting Mirapex a couple months ago I gradually lost 8 pounds of fluid retention that I had been bothered with.

My neurologist insists that GABA does not cross the blood brain barrier and I insist that it has pronounced effects! I found a chiropractor that said my BBB must be damaged.

I have a scientist acquaintance that told me the same, that GABA orally will not make it to the brain and when I took 250 mg GABA alone it seemed to be nothing. However, when I took it along with my Sinemet I perceived better effect on my PD symptoms from that combo than Sinemet alone. Then much later I discovered GABA-plex and tried that instead of GABA with my Sinemet and voila a mood calming effect, in addition to helping with the shakes. I took that as another improvement and have continued. Recently, really 3 days ago, I began to increase the GABA from 250mgx4/day to 500mgx4/day, still taking it along with Sinemet. I haven't noticed much change yet but I plan to go on for a few more days before making any change such as returning to previous dose or increasing. Rejuvem posted that they read where NIH published that 3000-4000 was an optimal dose for anti-depressant effect. I don't know, everybody has different needs. Just never giving up hope to find better symptom control in any way I can get it.

How I take GABA is certainly individualized. I am no scientist and I just tried it a few months ago and found it helped my mood and have been taking it in I think relatively small doses (250 mg) along with Sinemet. I recently discovered that if I take anymore than that at the same time as Sinemet I get less control of my PD symptoms out of the Sinemet. So I started taking an extra 250 mg capsule of GABA between Sinemet doses if I feel like it (a couple of times a day). just to help with my mood. So I am taking 1500 mg/day during waking hours. If I wake in the middle of the night to relieve myself and feel tremor and stiffening of my legs I will take more Sinemet and another 250 mg GABA. If that helps you, your welcome.
