I have been taking my peak flows for over a year now morning and night when I first started my best peak flow was 550 and was quite often near it. Now my peak flow is around 400-450. Do I now move my peak flow to a new best?

No, because 550 was your best and should still be regarded as optimal for you. I would say the new PF's are lower compared to that, but do you feel fine? if so then don't worry but I'm the same this year down from 680-740 to around 570-620 but not worried as I feel fine, advice is to keep an eye on it and symptoms.
