cancer, coronary artery disease... melatonin question

Healthunlocked repeatedly keeps deleting my posts in the middle of typing, so ill keep this brief, in hopes that it won't get deleted as well. I had just typed a long post but it got deleted twice... This has happened over a dozen times the past few months. This time, I am saving my post after every sentence.

In short - I have coronary artery disease. I exercise, I'm lean and muscle toned, eat very healthy etc. but it's a genetic issue.

I want to know your thoughts. does melatonin help with coronary artery disease, or make it worse? I've read studies and articles saying it helps but this article seems to say that melatonin makes it worse. I'm I reading this medical-speak correctly, or not? Your thoughts?

FYI, my platelet count has been a lot lower since around the time I started taking 3mg of melatonin daily, so they might be related.

"Earlier reports have suggested inhibitory effects of melatonin on agonist-induced platelet aggregation and secretion [12]. However, we show here that, melatonin, at higher concentration reachable by therapeutic doses, is capable of significantly raising platelet intracellular calcium even in absence of any agonist, while it synergizes the effect of platelet agonists. Recently, it has been shown that exogenous melatonin elevates [Ca2+] in pancreatic AR42 Jcells, which facilitates release of digestive enzymes downstream of physiological secretagogues [24]. We show here that, melatonin-induced rise in [Ca2+]iin platelets was not prevented by cell-permeable antagonists of IP3or PLC, suggestive of signaling mechanism unrelated to IP3-PLC axis. Our results supported that melatonin released calcium from the mitochondrial source that is known to play minor role in intracellular calcium homeostasis in platelets. As platelet is a critical player in arterial thrombosis leading to ischemic pathologies like coronary heart disease and stroke, and intracellular calcium being a major determinant of platelet reactivity, any therapeutic use of melatonin should be monitored under intense caution."

Maybe the key words in this article are, "therapeutic/higher concentration use of melatonin". I found this on a different site:

I didnt intend to insinuate that Darryl is deleting them, just that something might be wrong with the software, which is causing posts to accidentally delete while someone is typing them out.

There may be something going on with the software. My spacebar does not work when typing on this forum . I have to go back and space out each word.Only on my laptop and doesn't happen anywhere else.

what’s happens to me, is that i will be typing and then the screen will just exit and the message is wiped out, but I’ll still be on their website, which is how i know that is caused by healthiness unlocked software. It seems to happen not often, right after i hit the space bar but not always.

Why not just write in Word/WordPerfect/etc., then copy and paste to the forum?

me too. Many studies contradict each other., saying the opposite about affects on the platelet count.

I take 10 mg of melatonin daily to help me sleep, and I have low platelets (116). However, I’m skeptical of any cause and effect. Rather, I think my low platelets are due to the whole pelvic SR I had 4 years ago. Furthermore, I had a heart blockage 3 years ago and had 3 stents put in, so it’s actually good that my platelets are low, within reason, to avoid clotting around the stents. I’m not sure how any of that relates to your situation, if at all, except we share an interest in PC, heart disease and melatonin.

i had radiation too but it failed to do anything except leak into my body and cause the cancer to start growing at a rate 20 times faster than before the radiation treatment. I too got three stents put in the same year 2016. I was the second leading scorer on my hockey team, asa defenseman, and ran five minute miles regularly. Shit comes out of nowhere. Studies show that Heart disease and prostate cancer are frequently related.

My platelets for four years were around 150ish. They started going down around 100 or lower, this year, although, a test last month was at 155 but not it’s back to 110. I started melatonin around the same time the platelet counts began dropping.

My platelets are also low since RT to pelvic 10 months ago - around 110-120. My doctor says I’m ok with that as long as the value is higher than 30.

Wrt your last question, ie kidneys, the specified dose is huge. Even for a small (110 lb) person, that's 500mg per day, and you are taking 3.

that’s probably the issue. Just like with almost all supplements, low dose is probably helpful, high dose frequently detrimental.

Have you tried just composing what you have to say in a file using your favorite word-processing software, then just cut and paste the whole thing into your post?

Most studies seem to be positive towards melatonin. Here’s a trial on humans, although they are also diabetic.

Background & aims: Melatonin may benefit diabetic people with coronary heart disease (CHD) through its beneficial effects on biomarkers of oxidative stress and cardio-metabolic risk. This investigation evaluated the effects of melatonin administration on metabolic status in diabetic patients with CHD.Methods: This randomized, double-blind, placebo-controlled trial was conducted and involved 60 diabetic patients with CHD. Subjects were randomly allocated into two groups to receive either 10 mg melatonin (2 melatonin capsules, 5 mg each) (n = 30) or placebo (n = 30) once a day for 12 weeks.

Results: Compared with the placebo, melatonin supplementation resulted in significant increases in plasma glutathione (GSH) (+64.7 ± 105.7 vs. -11.1 ± 137.6 μmol/L, P = 0.02) and nitric oxide (NO) (+0.9 ± 4.7 vs. -3.3 ± 9.6 μmol/L, P = 0.03), and significant decreases in malondialdehyde (MDA) (-0.2 ± 0.3 vs. +0.1 ± 0.5 μmol/L, P = 0.007), protein carbonyl (PCO) (-0.12 ± 0.08 vs. +0.03 ± 0.07 mmol/mg protein, P < 0.001) and serum high sensitivity C-reactive protein (hs-CRP) levels (-1463.3 ± 2153.8 vs. +122.9 ± 1230.4 ng/mL, P = 0.001). In addition, taking melatonin, compared with the placebo, significantly reduced fasting plasma glucose (-29.4 ± 49.0 vs. -5.5 ± 32.4 mg/dL, P = 0.03), serum insulin concentrations (-2.2 ± 4.1 vs. +0.7 ± 4.2 μIU/mL, P = 0.008), homeostasis model of assessment-estimated insulin resistance (-1.0 ± 2.2 vs. +0.01 ± 1.6, P = 0.04), total-/HDL-cholesterol ratio (-0.18 ± 0.38 vs. +0.03 ± 0.35, P = 0.02) and systolic (-4.3 ± 9.6 vs. +1.0 ± 7.5 mmHg, P = 0.01) and diastolic blood pressure (-2.8 ± 7.3 vs. +0.1 ± 3.6 mmHg, P = 0.04). Melatonin treatment also significantly increased quantitative insulin sensitivity check index (+0.006 ± 0.01 vs. -0.004 ± 0.01, P = 0.01) and serum HDL-cholesterol (+2.6 ± 5.5 vs. -0.01 ± 4.4 mg/dL, P = 0.04). Supplementation with melatonin had no significant effect on other metabolic parameters.

Conclusions: Overall, melatonin intake for 12 weeks to diabetic patients with CHD had beneficial effects on plasma GSH, NO, MDA, PCO, serum hs-CRP levels, glycemic control, HDL-cholesterol, total-/HDL-cholesterol ratio, blood pressures and parameters of mental health.

thanks darryl, hospitals know the buzzwords, and they advertise cardio-oncology but then the doctors aren't actually trained in this specialty. I have an appt this month with someone who is supposed to understand this field, but we'll see how legit they really are, and whether they know about the cardio and the oncology, once I start asking questions.
