In regards to the clinical trial with this drug and colorectal cancer was so amazing . May this be used for prostate cancer as well? Is this similar to Keytruda?

Yes it could be used if the cancer has mutations such as MSI-H or d MMR.

These mutations are infrequent in PC.

How about CDK12

It may offer some control of the cancer in a minority of these patients.

No.

It seems the genes involved in MMR are :

"" MMR machinery in humans has 8 genes that code for its components: hMSH2, hMSH3, hMSH5, and hMSH6, while MutL homologs are hMLH1, hPMS1 (hMLH2), hMLH3, hPMS2 (hMLH4) (Lipkin et al., 2000; Clark et al., 2013; Amaral-Silva et al., 2017).""

CDK12 gene is related to:

""CDK12 regulates the transcription of long DNA repair genes (such as BRCA1,BRCA2,ATR,ATM,Fanconi anaemia), predominantly those involved in the HR repair pathway ""

""Homologous recombination (HR) is a molecular pathway involved in a multitude of processes, from the generation of genetic diversity to DNA repair and replication. HR provides a mechanism for the accurate repair of DNA double-strand breaks, protecting cells from chromosomal aberrations such as those seen in cancer.""

yes, both bind to the PD-1 receptor in the T cells.

I read (in a U.K. newspaper) that MSK are planning a trial of Dostarlimab to include prostate cancer patients among others cancer types. However the paper provided no further details. I expect that there’s more information in the U.S.

Newspapers tend to be very upbeat about potential new cancer treatments, there’s no harm in asking MSK, but please manage your expectations; the reported colorectal study result had a small number of participants. Also there was no mention of the outcomes for endometrial cancer patients for whom the drug was initially targeted.

Like most cancer patients, I too want this to be a wonder drug capable of curing all solid tumor cancers at any stage of progression, but I caution it’s very early and too soon to invest my precious hope only to have it dashed.

I wish you and your husband the very best.

Hugh
