Anyone on Magnitude clinical Trial (Niraparib)?

I am on the magnitude trial. Only a month in, no definitive results, few side effects (except the mother of all constipations) For those who don't know, Magnitude is a phase3 (large numbers, late stage of getting FDA approval) It combines aberaterone with niraparib, which is a PARP inhibitor that is considerably stronger than Olarparib. It is already in use for breast cancers. It is run by Janssen/Johnson and Johnson. Big big pharma. They make abi too so it is a pretty obvious strategy to extend the exclusive period for abi. Only for BRCA mutations. I am BRCA 2.
I am in Sydney Australia and go to the adventist hospital (The SAN). My oncologist is head researcher in the trial here. There are 3 arms to the the Magnitude trial. The first 2 were firstly patients with no genetic selection, randon blind assignment to active pill (still taking Aberaterone). The 2nd arm is selected for BRCA mutations. Again, 50% don't get the niraparib. The 3rd arm is open for BRCA - everyone gets Abi+niraparib. I am in that arm. The first month has been a pretty concentrated lot of tests, examinations etc. That gets less as time goes on. So far so good.
Hi "kaptank", thanks for the reply. I'm on the arm with BRCA but blind so I get abi + 50% chance of niraparib. I'm 4 months in, PSA has dropped from 11 to 4. I have a doctor appt tomorrow to see the results of my latest scans to see if the tumors on spine has stopped multiplying. Is your niraparib pill separate or combined with the abi? Where did you read that Niraparib was considerably stronger than Olarparib? Good luck on your journey.
The niraparib is in a combined pill with abi. I don't recall where I read that it was more powerful (ie flattened PARPs/took them out of action) than olarparib, but I do recall that the more recently developed talazoparib was about 100 times more potent than olaparib and rucaparib, with niraparib somewhere in the middle between them. Google naraparib vs talazoparib and you get an an unattributed statement to that effect first up. Good luck with your scans and let us know.
