Missed two days of my levo as had to wait for pharmacy.
In my opinion no , I have beer on Thyroxine for over 50 years and am now on strength 175 mg. Daily. I have had this situation and not been affected.
if you miss a couple of days of your thyroid medication then yes, you can feel very unwell and previous symptoms can appear. Heavy, aching legs and tiredness are definitely hypothyroid symptoms. Hopefully these symptoms will disappear now your pharmacy has your medication in stock/ when you are back to usual dosage.
I always have some spare thyroid medication and never wait until I’m down to the last pill strip before re ordering. It would scare me too much to run dry!
I used to do the same - request my next prescription when I got halfway through my current packet - so I was putting the prescription in two weeks in advance of the next one. Then one month I was told by the surgery not to put my prescription in earlier than one week before I need it - otherwise they would (and they did) ignore the request. But now the pharmacy is not able to deliver my next packet before I’ve run out of the last one. Good job I have a pack in hand. I suggest Scruffy1491 asks GP to give her an extra pack to prevent problems.

I can tell by about 2pm if i have forgotten my morning dose of Levo ..its subtle , but i can tell.
i definitely know about it if i miss a whole day , missing 2 days would leave me feeling very 'off' indeed .
Some poeple wouldn't feel much difference at all until they had missed several days, and wouldn't get actual 'problems' until they had missed a few weeks .

The two hypos I know best are affected very quickly - hours. But others seem not to notice anything for much longer.
I think if you are in one camp, it is difficult to imagine being one of the other lot!
many yrs ago (when i wasn't so 'tuned in'), the first consequence i noticed of forgetting a mornings dose was that i would be disorganised and invariably forget the next mornings dose too... i'd usually figure out what had happened on the second afternoon.
I've never 'not noticed' for longer than that .
Nowadays the first consequence i notice is on the first day ~ i'll start rabbiting 'too much' to anyone passing at about 11am, not wanting to stop for lunch , wanting a fag instead , feeling a bit "adrenaline-y "
if I get a bad migraine, then I can’t take my thyroxine or it makes the migraine worse. I have to take it very late once the migraine subsides.
I recently had a meeting with my endo. Told him about the migraines. He said I should take my thyroxine dose as soon as I feel ok to, and then continue with my doses as normal, then I haven’t missed out on any.
I haven’t done this yet. I’m very sensitive to doses. I usually make up the missed dose gradually over a week.
