Trastuzumab Deruxtecan Extends Survival Over Chemotherapy for Patients With HER2-Positive Metastatic Breast Cancer
The trial included 608 patients randomly assigned (2:1) to either T-DXd or TPC (trastuzumab plus capecitabine or
lapatinib
plus capecitabine). The researchers found that patients had a median of two prior lines of systemic therapy (excluding hormone therapy) in the metastatic setting.

Just in time for Halloween: Not so fast when zapping all those senescent Zombie Cells, as some might actually be your friends.
As with most new insights into the complexities of the human organism, things usually prove to be more complicated that they first seem. Even a cursory look at genetics reveals that in very short order. And for those hoping to use Occam's Razor to make such organic complexity seem simple, it may be more

? Experience w/ taking Hydroxychloroquine & ADT simultaneously? If so, issues w/ your heart or other side effects? Clinical Trial or Nubeqa?
I've been considering doing a clinical trial or just starting on Nubeqa. The trial includes a pretty high dosage of hydroxychloroquine (HCQ), along with Metformin and Sirolimus. I'm trying to figure out if the trial/HCQ has a significant chance of causing heart disruptions/issues. I am cautious because

Oxidative Stress (ROS), Sleep and the Gut Microbiome: Staying Alive
I am just back from a challenging (humbling) multiday backpack in the High Sierras of California. Very beautiful but drought conditions, dry stream water sources made it more difficult. Here is a pre-note on my own regimen: After completing my two phase radiation treatments for oligometastatic PC:

Help and advice please.
Hello again everyone. I have just started on Tagisso which my doctor said is unlikely to work because my tumour doesn't have the right genetic 'make up'. But I live in hope, what else is there. He also said that if it didn't work there was nothing left. Over the past 2 years I have had Afatinib, immunotherapy

Senolytic Regimens for Treatment-Emergent Senscent Cells
Previously, I posted on the topic of cellular senescence, "zomby-like" cells that cannot divide but also do not die. They can become SASP cells, Senscent-Associated Secretory Phenotypes, that can promote the growth and progression of processes in other cells, including PC cells by secreting pro-inflammatory

Dasatinib
I was interested to see in the latest from the Mayo: "
Antidiabetic Effects of the Senolytic Agent Dasatinib
" [1] Many men with PCa are pre-diabetic at diagnosis, with some of the symptons of the metabolic syndrome [MetS]. Pre-diabetics do not necessarily progress to diabetes, but androgen deprivation

Senolytic combo Dasatinib + Quercetin
Nalakrats has in two major posts shared fascinating insights on senescence and senolytics, drugs that selectively clear senescent cells. I am most grateful to him for this, which set me off on a journey the results of which I sum up in this post. Senescence has also recently been well reported on by

A large injection of hope required. Please.
I was diagnosed with stage 4 lung cancer in early 2020 and was given the targeted therapy Afatinib. This was initially most effective but gradually stopped working and so the cancer spread further in the lungs. I then had five consecutive radiotherapy sessions which reduced the size of the main tumour
