Oh Dear. I realise its the title of the program, but if we can all go easy on posts talking about miracle cures

This is a study of particular interest to me (see my remark a few minutes ago to PDConscience on his thread). In particular GDNF-7

And best I recall the trials, they were a success in terms of developing an effective delivery mechanism, but a failure in terms of the "drug" (its not a drug, it's a natural protein - the one John Pepper gets so excited about) having any beneficial effects.

So, the current idea, still at lab mouse level (remember PWP are NOT mice) is to modify the compound to include miR7. The problem appears to be that in living PWP the uptake of GDNF in the neurons we want to heal is blocked by - you guessed it - excess alpha synuclein (note not fibrils or lewy bodies, but "spare" protein). This turns out to be due to a lack of a microRNA - number 7, which is also expressed in healthy cells and binds to the alpha synuclein to stop it blocking the GDNF uptake.

Anyway, I'm sure the program will explain more and better, but don't get your hopes up. The miracle hasn't happened yet (as far as I am aware).

Maybe GDNF and my antibody BIIB054?... ?

Yeah. Problem is gdnf from exercise is produced in glial cells (on nerve sheathes) and doesn't cross the blood brain barrier. To the extent that any is produced in the brain by those mechanisms it is ineffective at healing neurones in the substantia Nigra of people due to inadequate mir 7 leading to alpha synuclein blocking the receptors. At Bristol they were pumping the stuff in

On lab mice with artificially killed dopamine neurones it regrew new pukka full function neurons

But lab mice are not pwp and they don't have a defective mir7 issue.

If a keto diet produced gdnf in the right neurons then half this forum would have brand new dopamine neurons and be cured of PD.

I thought it was a consensus view that exercise produces GDNF for the brain, but not so, huh? What about stuff we can ingest?

Ibogaine in micro doses has been proven to increase GDNF in the brain.

Thanks. I don't know anything about that stuff, except I thought it might be potentially harmful. Do you take it?

I took it for six months and have experienced a sustained improvement in my symptoms. I am currently on a hiatus from it. I plan on taking another course of it. I do think it caused me to have low blood pressure while I was taking it.

Ingested gdnf will not cross the blood brain barrier. This BBC program which is the subject of this post is about a trial program at Bristol university to administer gdnf to regrow dopamine neurons specifically.

In lab mice,who didn't have PD but had their substantia Nigra neurons toxically destroyed,they injected gdnf and grew perfect new neurons which great proper tendrils into other parts of the brain .

Tried it in pwp.didnt work. developed a really high tech permanent brain catheter to allow easy clinical administration of gdnf direct to the SN in the brain. Didn't work.

Latest thinking is that mir7 is to blame and they are currently testing on different mice to see if it's worth moving to human trials.

But a tiny bit of extra gdnf even in the brain is not going to deliver what this research is looking for. Maybe it has other benefits but it's not this project

The full results of the GDNF trial are due to be published on 27 February 2019.

The first half of the BBC2 doco is scheduled for 9pm on 28 February 2019.
