Five years into PD after diagnoses now. Quite recently something weird started to happen. The first C/L dose either take Lon time to kick-in or it does not kick-in at all. The second dose work like s charm. Food is not s factor here since I do intermittent fasting and my first meal is after lunch after the second dose. Dose remained the same for over a year. Any explanation for what is happening here?

If you are also taking a multivitamin or B complex that contains vitamin B6 within a couple of hours of taking your C/L that will inactivate it. See here for details: healthunlocked.com/cure-par...
Thanks park_bear! I have tried not take b6 or b vitamins before the first dose, taking it more than one hour before, and one hour after, always the same result! Is it safe to take b6 at night, or after the last dose before sleep without causing sleep problems? I'm thinking I might have b6 deficiency from consuming large dose of C/L and b6 is necessary for ldopa to dopamine conversion.
As far as I know, vitamin B6 does not cause sleep problems. If you cannot separate taking vitamin B6 from taking C/L by 2 hours, separate them by a meal, or wait until you're fully on from the last C/L dose.
As noted at the link, you should also be supplementing vitamin B2, and be using P5P for vitamin B6

People have been experiencing peripheral neuropathy not only in Australia but also in other countries.
You should consider how much B6 your are getting in your diet PLUS the B6 supplement….. maybe you are getting to much B6 and hence why you are having issues with your C/L medication.
If so, supplement with Riboflavin.

I recently had to start taking a slightly larger first dose. I was having the same issue. I know eventually the carbidopa Levodopa quits working. I'm wondering if this is the beginning.
I have this sometimes. Usually either after a few days of eating certain foods (animal protein, legumes and possibly tomato). Recently it happened when I switched to pomegranate juice for the morning drink (like you I skip breakfast)Did you change what you drink or any other food or supplement, considering the day before?
Interesting. I have heard of C/L doses not working during a medical induction interview with my movement disorder specialist. Its s real phenomenon. I don't know much more about it but I would talk to a movement disorder specialist. Mine is at a big city hospital. My local neurologist is great but sees more seizure disorders than PD.
My theory is when you are still producing some of your own dopamine it peaks at midnight and is at a lowest at midday. Any doses of levodopa you are on are overlayed on your natural cycle.
As the years go on that overnight replenishing decreases so now you are going say 8 hours with no natural boost and no dose of levodopa so you are at a very low point in the morning. You need an extra dose first thing to bring baseline bloods back up to the level they used to be naturally first thing.
Try taking a couple of doses 2 hours apart first thing or
1.5 dose first thing, or
an extra 1/2 dose between the first 2 doses in the morning.
See if any of those help. Try them each for a few days to give it a chance until you find one that works.
