I strongly believe, and have for a very long time that Parkinson is caused by a virus. It must be a slow growing virus and judging by covid-19 where some people are not affected by it, it must be the same with Parkinson's. Where some people even if they are exposed to the virus don't get Parkinson's. What are your thoughts on it? That is why some of the families have a lot of Parkinson's. Because it must be contagious if you are susceptible to it.
I think more likely it is caused by having certain bad combinations of gene mutations that interfere with breakdown and formation of neurotransmitters. These may be turned on by viruses or other toxins. People in the same family will have some of the same mutations but some may have different environmental exposures so the genes may not be switched on.
i don't think genetic (at list human) is the way to go. we have 24.000 genetic units, rice has 40.000 and our microbiota 4.000.000 so .... we are our microbiota.
a simple gut dysbiosis (wrong microbiota) can generate lack of aminoacids (like tyrosina or tryptofano) that are fundametal for brain function. With lack of "fuel" the neurons can be switched off o disconnected to save resources and/or because useless.
Meds change (and cronicise) situation from "struggle for more levodopa" to "all this goodness of leovdopa" due to the huge external supply. Neurons stop producing levodopa and patient become dipendent more and more on external supply.
Quantity is huge and neurons are saturated with spikes of levodopa that is metabolized in toxic elements (es. 3-O-MD = tri-ossi-methildopa) therefore becomes less and less sensible and standard solution is to increase the meds (big mistake).
Carbidopa zeros B6 and this cause a long term scarsity in the deep brain that damages the neurons and decrease the capability of neurons to manage spikes.
I believe in scientific evidence. If the slides from 14 pwp show viruses in different stages of development and staining points to enterovirus I think it deserves serious attention. Given 0 yes zero progress in disease modifying treatments I think that further research into virus origin of pd is imperative. Look at polio which is caused by a enterovirus and you see many interesting parallels with pd. I wouldn't be surprised that genetic defects contribute to immune system defects that makes us more likely to develop pd
