Time to steady state of nebivol calling all mathematicians

Assuming a starting dose of 62..5 mgs once a day. I gather half life is usually 12 hours though in poor metabolisers up to 30 hours

Not idle curiosity. 5 days ago I stopped N. 2.5 mgs once daily with startling benefit in general well being and exercise tolerance.

Here's the rub. I became aware yesterday that my heart rate was 116 regular at rest. I thought atrial flutter which I’ve had before. I’m usually permanent AF. At all times in the past weekI have felt fine. Kardia 1 said tachycardia.

Yesterday I restarted 62.5 mgs. My question is how long should I wait before considering a further dose increase?

The half life makes a significant difference. I'll give 2 examples using 1/2 lives of 12 and 24 hours (because this makes the maths easy to follow).

I assume you mean 6.25mg Nebivolol daily dose as 62.5mg appears very high?

Daily dose 6.25mg, 1/2 life of 12 hours means that 3/4 of whatever is in the body is excreted by the time the next does is taken. So peak levels are

Day 1 - 6.25mg

Day 2 - 7.812mg (=1/4 of original 6.25mg remains + new dose of 6.25mg)

Day 3 - 8.203mg (=1/4 of 78.12mg remains + new dose of 6.25mg)

Day 4 - 8.300mg

Day 5 - 8.325mg

Day 6 - 8.331mg

Day 7 - 8.332mg

Day 8 - 8.333mg

Pretty stable peak level of about 8.3mg after about 4 days

Daily dose 6.25mg, 1/2 life of 24 hours means that 1/2 of whatever is in the body is excreted by the time the next does is taken. So peak levels are

Day 1 - 6.25mg

Day 2 - 9.375mg (=1/2 of original 6.25mg remains + new dose of 6.25mg)

Day 3 - 10.937mg (=1/2 of 9.375mg remains + new dose of 6.25mg)

Day 4 - 11,718mg

Day 5 - 12.109mg

Day 6 - 12.304mg

Day 7 - 12.402mg

Day 8 - 12.451mg

Pretty stable peak level of about 12.4mg after about 8 days

NICE (UK National Institute for Health and Care Excellence) recommend allowing 1-2 weeks between dose changes.
