Hello all.

My dad has just been matched and accepted on to the ODM-208 trial. Does anybody have any experience/knowledge of this? Thanks in advance.

I was on this trial. My PC progressed very slowly while I was on the trial but I felt great while on it for about 5 months until my progression was too much and Lu-177 had become available again so I left trial for Lu-177.

The extra check up's such as EKG's etc. along with extensive blood lab work ups was an extra benefit.

The effect on adrenal gland and others means taking steroids and the trial staff questioning for and looking at blood labs for signs of Adrenal Insufficiency. It is necessary to carry a emergency pack they give you to inject steroid if you have si gns of Adrenal Insufficiency or Adrenal Crisis. I never experienced that.

I think ODM-208 is very promising. I think of it as a supercharged Zytiga (Abiraterone).

They will also check for a mutation which in the phase 1 trial seemed to effect the positive or negative response to the trial drug but they had few subjects/patients to clearly confirm that so the phase 2 trial is letting in a larger number to try to also look at the response in leu of mutation or no mutation. (somatic point mutations)

Thanks for this, very interesting. I naively assumed my dad would have the mutation as he was told he was matched to the trial but not necessarily if the phase 2 is still comparing with/without mutation?

It was a little confusing. I think they are looking at both groups. But as a phase 2 also looking at chosen dose and SE's etc.

The mutation was a little confusing. I think those without the mutation they looked for did better.

The trial Dr's are very thorough and take time at appointments. I'm sure they can answer that more accurately. My brain is at about 40% efficiency lol.

I was confused about that last May and got it straight in my head and then forgot again.

First let me correct a reply I made to you a month ago. Those with the mutation did better in the phase 1 trial ( I informed you the other way around).

From phase 1 trial:

Overall 12/36 (33%) evaluable patients achieved a PSA decline of ≥50%. In evaluable patients with AR LBD mutation 10/15 (67%) achieved a PSA decline of ≥50%. Clinical improvement in symptoms such as pain was also observed in some men.

So onto you current question. I did not have the mutation. Which seems odd given my PC's hormone resistant state but anyway it is how it is.

I also noticed looking more closely at my medical records that when I was admitted to the hospital ER diagnosis of Adrenal Crises was listed. However I was quite sick with and remain sick 3 or 4 days out of 7 with Cyclical Vomiting Syndrome so its a little dicey I think giving the diagnosis of Adrenal Crises. Another diagnoses listed was starvation (more fitting given vomiting and no appetite. I definitely needed IV's that I got. I've adjusted a bit to the vomiting syndrome and have been able to nourish myself well enough but I'm not gaining back the 50 pounds I lost. Just staying even.

Anyway enough babbling, possibly venting by me. Again I didnt have the mutation.

I hope he goes on the trial. I see promise in it and the care and observation given by the Clinical Trial Dr's and staff ensures a high degree of safety which is designed also into the trial.

I have already finished about three or four weeks ago. I had a great response. After my fourth session I only had one small faint area on my scan. So I am very grateful to God. It’s worked well for me. It was just nice to hear, someone else having a good response. I only wished it worked for everyone. I am glad your response is good. And only gets better as you finish. Thanks so much for responding.
