Has anyone with the PIK3CA mutation heard about a new CDK 4/6 inhibitor called Aplelisib? Has anyone been in a trial? My oncologist mentioned it as a possible alternative to Ibrance that is more targeted to this particular gene mutation. It is apparently waiting for final FDA approval. If I start it I'll keep you all posted!

New CDK 4/6 inhibitor for PIK3CA mutation?

Hi Lynn! Alpelisib was approved by the FDA in May so it should be available now or very soon. Worth asking your oncologist for more info, especially if you have the mutation. Ibrance/Faslodex didn't work for me (liver mets increased) so Alpelisib was one of five options presented to me in May as a next line of treatment. Based on my oncologist's opinion and the second opinion I got at Dana Farber, we decided to wait until it has more of a track record, is easily available, and perhaps can be paired with Letrozole. (right now it's only approved for use with Faslodex, which didn't seem to work for me.) I needed to start something new right away so went with Xeloda, an oral chemo that has flexible dosing and so far few side effects. My onc did say that I could try Alpelisib after Xeloda too, and by then (hopefully at least a year or two from now) they'll have more dosing and combo options. I'd love to hear your oncologist's opinion about it!
