Why should I not eat Navel Oranges or Grapefruit while using Olparib?

Always looking for the broader information found on this site.

Anyone know why we are not allowed to eat certain citrus foods while on Olaparib?

And why just Navel Oranges and not Valencia Oranges, for example?

What about Tangerines, Clementines, etc? What does a Navel Orange have that a Valencia oranges does not have?

Any input appreciated.

Grapefruit or pommelo has a chemical that blocks an enzyme (CYP 3A) that is required to metabolize some drugs, like PARP inhibitors; so, the drugs can build up to toxic levels. Some citrus, like sweet orange or tangelos, are created as a cross between grapefruit or pommelo and mandarines. So the less like a pommelo the hybrid citrus is, the less one has to worry about it inhibiting CYP 3A.

"Because degarelix does not seem to interact with the cytochrome P450 enzyme system as substrate, inhibitor, or inducer and does not show any interaction with hepatic and renal uptake and efflux transporters, the risk for pharmacokinetic drug-drug interactions with this compound is highly unlikely."

Best to check any other supplements you may be taking. Probably no harm but I would run it by your treatment team.

Depending on you medication anything else you put in your body must not interact to lessen the treatment effect.

As an example, I was on a PARP inhibitor, I experienced stiffness in some joints.

Took a joint supplement which had a high concentration of Turmeric in it.

The turmeric has a negative effect on the CYP3A4 enzyme (WHO KNEW!!!)

It turns out that in the literature that the trial gave me it stated....

"Do not take anything that interacts with CYP3A4".

Caught it in time by checking the MSK app about herbs. (Definitely worth downloading to your phone.)

The joint pain went away, but that was a close call.

It makes you wonder how many guys are not having a response to PARPi because they are eating something which srews with the drug?
