Are there other brothers out there with the CHK2 gene mutation? How is it being treated?

Yea I'm Chek2 I've been on ADT therapy now going on three years. Generic Zytiga, Eligard, and Xgeva for bone mets. I also had Provenge infusions a year ago .My PSA has been 0 1 since first month treatment. My Doctor said this gene mostly is inherited so my children were all advised to be checked, it is also associated with the chance for other Cancers. I've responded well to my line of treatment so far st good. Leo

I’m in that club, I’ve been on Lupron since dx back in 2014, Xtandi for 5+ years, Avodart 5+ years, metformin 5+ years (Snuffy Myers protocol). I’ve also had my prostate and several lymph nodes radiated back in 2014 and chemo per CHAARTED in 2015.

So far after 6+ years my stage 4 cancer remains undetectable (knock, knock).

My son also carries that mutation so he knows to be on the lookout early, fortunately my daughter does not. I now see Dr. Sartor at Tulane who did genetic testing on me and made it available for my family.

thanks, my son also carries the gene. On the lookout for possible treatment like Lymparza.

I think for the most part my Doctor knows my options and he has promised me he would try his best to have me watch my Grandkids grow up

According to Sartor none of the current PARP inhibitors are effective against CHK2. At least that’s my recollection of our conversation.

Not on their own but they are having some success with Lynparza paired with Keytruda. I hope that is true.

There is a clinical trial combining lynparza and Keytruda for those with that mutation. I just started this last week at MD Anderson. You may check and see if there are any hospitals near you that are involved in it.

I have CHEK2c1100 deletion, diagnosed with Invitae blood test. G9, 12/12 cores positive with 10% G9, 80% G7. I have had whole pelvis IMRT, HDR Brachytherapy with hyperthermia, spot SBRT to 3 <1cm indeterminate bone lesions, Lupron x 19 months and ongoing, Casodex x 4 months at first (liver toxicity), Erleada x 8 months and ongoing. PSA <0.1 since conclusion of radiation treatment phase 15 months ago, other labs stable and normal so far.

Thanks for replying. You are the 5th, so that makes 6 of us with this "rare" gene mutation. I'm fighting the monster for nearly 10 years since dx at age 72. G7(4+3), Radiation to prostate good for 4 years. Have gone through all the usual stuff and finished Taxotere last January. only on Lupron with psa rising, but, scans not showing much activity. Two guys might be starting Lynparza.
