Torisel for PTEN loss

Does anyone have information or experience with the use of a medication called Torisel (Temsirolimus) in APC in those patients that have a loss of PTEN?

Temsirolimus is a one of several drugs called mTOR inhibitors. Sirolimus, Evorolimus, Rapamycin, and other novel mTOR inhibitors have been tried as well. In general, they are highly toxic - which limits their use, and they don't do much - more if there is PTEN loss. Some combinations have been tried, but results have been underwhelming. Much more promising and further along in testing for PTEN loss (which is very common- half of men with mCRPC have it) is ipatasertib:
