I have sharp pain along one calf whilst running and a burning sensation for ages after. I am currently repeating week one until the pain gets less distracting. Will this fade or do I need to be concerned about claudication?

Minor aches and pains are normal when you first start the programme. Make sure you stretch after your runs

and consider taking two rest days between runs rather than one.
A burning pain is not a normal symptom. If you have a sharp pain you need to see a physio. Calf strain needs time to heal or you could put yourself out of action for a longer period of time.
