I am doing a few hours as a trial at a local cafe, it's quite small but also popular. The manager is slight bossy and intimidating (which I'm sure she doesn't mean to be).
I'm having anxiety just thinking about it, what if I hate it but I'm missing out as I might enjoy it a few months on? How do you know you'll be comfortable somewhere?

You will never really know if you will be comfortable in anything you do in the future. Focus on the now. You must trust your gut, try not to take it all too seriously, learn all you can and try laugh as much as you can along the way too. I hope that helps.
If you don`t start this job, you`ll feel bad , as you`ll never know how you would have coped. What`s the worst that can happen ? The manager wouldn`t have employed you, surely, if she thought you weren`t capable. Try visualising the day at work, with everything going brilliantly. Even if you don`t believe it consciously, your inner thoughts are lined up and ready for a good day. So, don`t worry, above all, stay calm, as this fear will cloud your common sense....and be truthful to the manager if you can`t do something, or are nervous- I`m sure she will understand. A new job is, understandably, a little daunting.
