Would like to find out if anyone has had olaparib before .. How effective is this drug? Would anyone know if this is the next line of treatment if cabaxitazel is rendered ineffective? Thank you.

Olaparib
Olaparib is very effective, but only in men who harbor the BRCA mutation. Have you been tested for that? Or does PCa or BCa run in your family?

There was a recent thread on the old Yahoo site, & one man reported using Olaparib & responding, as I recall. I doubt that I would respond. Not aware of any familial PCa/BCa.
The concept is quite neat. Cells with certain DNA damage, such as those with BRCA1 or BRCA2, can remain viable due to the repair activities of PARP. Olaparib is a PARP inhibitor. Cancer cells that are not dependant on PARP repair are not affected.
BRCA1/2 is quite nasty, affecting men at an earlier age & being quite aggressive. But affected cells turn out to be vulnerable. The Olaparib effect is like kicking away the crutch of a one-legged man.

I have the BRCA2 gene and my oncologist put me on olaparib and my PSA went down from 50 to 12 in 4 months and now is starting to rise. I've been on just about every treatment including Provenge and Xtandi and they have all failed after 4 or 5 months. I am presently on Lupron, Prolia, Lynparza(Olaparib) and casodex and my oncologist will try me on Zytiga.
