Lasofoxifene Combined With Abemaciclib

About to enter a drug trial through Mayo Clinic. Wondering if anyone has taken the combination of Lasofoxifene and Abemaciclib (Verenzio)? A year ago I had 11 months of great results with Faslodex and Ibrance which are in the same drug families so am hopeful I can tolerate and will be effective with my bone Mets. Thank you.

Hello,

I read studies about Lasofoxifene and how it is on the fast track of being approved as a new SERM treatment option. Where is the trial? Hoping for the best for you!

Mayo Clinic in Minnesota. Yes I've read good things as well. It will be my 4th line of treatment in 18 months so I'm hopeful I get some good results. Thanks!

Do you have the ESR1 mutation? What other treatments have you received?

I do have ESR1 mutation as well as Pic3ca mutation. After Ibrance/Faslodex for 11 months (3 scans NED then progression). Then did 2 months of picray with horrendous diarrhea and had to stop (during whick markers went way down), then 8 cycles of Gemzar. Mets and markers increased on chemo.

My wife entered the trial thee days ago, at Ohio State. She had a good run with Ibrance plus letrozole but then progressed after acquiring in ESR1 mutation. She then had a year with Verzenio plus fulvestrant. She showed progression in the liver in April She's now just started the Verzenio plus lasofoxifene trial. Verzenio in the past caused more diarrhea than Ibrance but was well tolerated. We're hopeful about the trial. Good luck!

How is your wife doing on the new combination of drugs? I had read lasofoxifene causes constipation and Verenzio diarrhea - maybe they balance each other out? A nice wish perhaps. By the way I think it's great you're on this forum sharing. Hope you continue to share since you're the only one I know of (on behalf of your wife) in this trial. Thanks.

My wife started the trial on June 10, just after the it began at Ohio State. She was on the drugs for a couple of days but then had to pause them because of unrelated health issues. The pills resumed last night, June 16. There's nothing to report so far; it's way too early. However, there's another trial at Ohio State, fulvestrant vs lasofoxifene, that opened earlier. From what I've heard, lasofoxifene seems to be a fairly friendly drug. I hope you're right about the off-setting BM effects of Verzenio and lasofoxifene. That occurred to me too. This trial seems biologically sensible to me, as opposed to a different treatment that would have sent a woman with an ESR1 mutation back to an aromatase inhibitor. I couldn't make sense of that.
