Do you think weight and height could have anything to do with Ibrance?

I have been noticing that some people seem to tolerate Ibrance better than others. I am on 125 mg. and have no trouble, other than low counts, but not low enough to delay taking it. When I first started they would delay one week due to low counts, but they do longer do that. I am wondering if weight and height could have anything to do with dosage tolerance. I am significantly overweight. I wonder if women who are at correct or under weight bodies might not tolerate higher doses as well as someone who is overweight. I am not referring to someone who is 20 pounds overweight. The fat might make it easier on the body. I have never seen anything about this but it seems like it could be a possibility. Any thoughts or knowledge? Blessings, Hannah

Not sure but other drugs are reduced depending on weight

I’m about 135 pounds and 5ft 8 ins...so quite slim and I couldn’t tolerate the high dose...on100 mg

I’m about two stone heavier than my ideal weight for my height so nowhere near slim but I’m only on 75 and that is only for two weeks on two weeks off. Most people take three weeks of tablets and have one week off . So I think we are getting closer to a conclusion that weight is not connected to dosage !

In medical terms I have no idea but I think palbo is about each individuals tolerance and immunity and that’s not something that can be measured prior to a clinical very well , I suspect. When my son was young , he was quite frail and had a reaction to paracetamol and the doctor did say that by body mass he should only have half a tablet but I don’t think palbo works that way - that’s just my opinion. Yes but the study says’ A] larger cohort is needed to elucidate the correlation ....’ . The sample used to correlate the results was too small so it’s inconclusive

It is an interesting thought. I have no idea. It could be related to the biology of the cancer and other issues. I was hoping that I need to eat more. Blessings Hannah

No. It was just food in general. It does seem that weight has nothing to do with dosage of Ibrance. The best to you. Blessings Hannah

That may mean that folks with less weight need less of the drug. May mean the more you weigh the more you need. Blessings Hannah

No,not in my case and from other forums boards I had been on, that doesn't appear to be an issue weight wise. Some people can handle it, some people can not. Some people felt the same on their week off of Ibrance, others felt very sick even after being on it for over a year.

I’m overweight. A lot overweight. I’m at 125 and yes I think that’s a big (no pun intended) part of the dosing. I am, however, trying to lose weight, so we’ll see how this goes. Doing it for lots of reasons, but want to give the meds a fair shot to work and I think body mass is part of it.

There was never any reports about weight being part of any factor in dosage. I am very overweight (obese, hate to admit it). I could not tolerate the Ibrance and my week off was the sickest I ever felt. I was on 125 mgs. Switched me to Verzenio at 150 mgs. (twice per day with no break). I stopped by second week, the side effects were too much, lowered to 100 mg. (again 2x a day) and felt the same as the 150 mgs. Could not even leave the house. Switched again to 50 mgs. (2x per day) and that was the trick. I feel normal. I no longer have to nap, I no longer have stomach issues or headaches. I hated the Ibrance and I was obese and could not handle it and yet now on Verzenio, lowest dosage and what a difference. I have almost no side effects and more importantly I do not have to take naps anymore. Big difference. I am actually (very slowly) losing weight bc now this drug allows me to go out and walk more and do more.

I am 55 and have tolerated 125mg Ibrance with no breaks for 20 months however I fall within ‘normal ‘ BMI weight levels (and was at the lower end of the scale when I first started Ibrance and I have a petite build ) , so I don’t fit these research findings ! I have always thought I have tolerated Ibrance well so far due to the fact that I have not previously had any chemo or radiotherapy prior to starting Ibrance and these can affect the bone marrow (where the white blood cells are made ) ? X

I’m on the 125 dosage and don’t have any issues, I’m 5 3 and weigh about 113 lbs, been on it since 2016.

I didn't tolerate it well and developed interstitial lung disease (ILD) while on 125 mg and I am far from slender! I wish the drug companies or FDA would publish incidence of various side effects! Ibrance and Afinitor and some of the even newer drugs are still so new that there isn't alot of data ......

Not true in my case. I am (hate this word) obese and was put on Ibrance at 125 mgs. First two weeks were great. Then third week I woke up and felt like somebody had beat me with a bat. I could barely get out of bed. Then came the week off that I was looking forward to. It was the sickest I ever feel. Although summer and humid and even having to turn my central air off and I could feel the stuffiness of my apt., I was freezing. Had pjs, heavy robe, three heavy blankets on and yet I was shivering and my teeth were chattering. I could barely get up, everything tasted like metal and then a common infection that I used to get precancer and usually cleared up with antibiotics got so much worse. Wound up having to have surgery in which they cut tunnels out of my upper thigh (the five weeks of antibiotics did nothing), I had holes in my leg. Recovery was supposed to take four weeks (I was off the Ibrance then). Took almost five full months and was a horror. I hated the Ibrance. All three weeks of my Ibrance were awful so I was taken off it. They switched me to Verzenio at 150 mgs. (that was awful and I stopped after two weeks), you take Verzenio twice a day with NO breaks. She then lowered me to 100 mgs. The same horrible nasty side effects that made me unable to leave my house. She then switched me to the lowest dose 50 mg. twice per day and that was the trick. No more naps, no more feeling sick...I almost feel normal. So even though I am extremely overweight (and was bed ridden for almost five months after the surgery) I gained even more weight. If I had not been on the Ibrance, the usual antibiotics would have worked on it before it got worse and I had to have surgery. I don't think weight has anything to do with as in my case.

I wonder if there has been any studies of weight and dosing for any of these drugs. Maybe it doesn’t matter because it seems people of all builds have very different tolerances and the art of it is trying different schedules and different doses until you can tolerate it and it is still effective. And if all else fails trying another drug. Anyhoo, I’m still watching my diet and trying to be more active because this extra weight is no good for anything. So far have lost a few pounds. No cookies. No cake or muffins 🥴

I’m lucky as I’m not keen on cakes biscuits etc

My downfall is ice cream...and I’ll indulge when I feel like it!!

I was on Ibrance for 6 months. It didn't work for me. I was on the highest dose along with Faslodex and Xgeva. I wear a size 6 petite and had no m problem with side effects other than dryness and losing my hair.

Interesting question. I would think weight would be one of the factors. But I think thete are so many more factors.

In my case I have never been able to tolerate drugs without side effects... even aspirin! Lol! Funny I would end up on Ibrance!

Hello! Without any medical knowledge, I would say that common sense would agree with your thinking. Medications are used by the entire body and a larger body uses the meds in a different way. At 5'2" I am considered overweight and I had downgraded to 100mg Ibrance because of struggles with neutropenia. I'm fine now but am taking the Ibrance on a 5 day on/2 day off schedule which avoids problems with neutropenia for me. But yes, Im pretty convinced that size makes a difference in metabolizing meds.
