OSI906 and Taxol

A friend of ours is starting a trial with weekly Taxol combined with a daily tablet called OSI906. Has anybody got some experience of this trial to date. Rgds Paul

Is your friend actually going to have this combination. Sometimes trials give a placebo to a proportion of the sample group. I found a reference to it in the Astellas Pharmaceutical US website at astellas.us/therapeutic/rnd...;

It states,

'OSI-906 is a potential first-in-class selective small-molecule, dual kinase inhibitor of both insulin-like growth factor-1 receptor (IGF-1R) and insulin receptor (IR). OSI-906 is currently in a Phase III clinical trial in adrenocortical carcinoma (ACC) and in Phase II clinical trials in lung and ovarian cancers.'

Looks as though your friend is receiving leading-edge drugs in the early trial stage. I'm looking forward to hearing how it goes for her.

She starts the treatment next Wednesday, I appreciate in many trials a percentage receive the placebo but she will be unaware if she has the real treatment or not. Sounds like a new ground breaking trial, I just wondered if anyone else has already been involved with this treatment. I have read up on the USA sites but nothing clear here in UK.

Hi Paul, I was on this 2nd phase trial at the Churchill Oxford from July 2012 to November 2012. Initially it worked very well for me reducing the tumour size and my CA125, it was end of October when my CA125 started to rise again and scan showed that although most of the tumours where still reducing I had thickening on my bowel so had to stop the trial. I had no adverse side effect from the trial drug at all, it was only the taxol that made me loose my hair again. I hope it works ok for your friend., If I can help anymore just ask.

She goes for prelim tests on monday ie ecg ect before starting, strange thing is her last scan showed thickening of her bowel, will this stop her proceeding do you think.

Hi again, I don't think that the thickening of the bowel would stop her from proceeding with the trial as everyone responds differently to each treatment. I am not entirely sure what the criteria for proceeding with the trial are apart from stage and type of the OC and what treatment you have previously had.

Thanks so much for posting up this information. It's so helpful to go armed with queries for the oncologists. I've been told if you ask for stuff you can often have it so am trying to keep a record of all the new stuff coming out.

Hi, I haven't found it too bad, although as the cycles go on I have found that the rash has got much worse all over my body although not always itchy. The other problem I have had is that it has taken it out on my bloods and magnesium levels, so far I have had 3 blood transfusions and I have to have magnesium influsions every week now as my body is not retaining it. The other side effect that I have had is that my bowels have been playing up in that they have been going into spasms which can be very painful and very noisey, this seems to have got worse as the treatment has gone on. I have found this all tolerable if not particularly very nice. Am hoping that bloods and magnesium and bowels will sort themselves out after I finish this course. Let me know how you get on with it.

Dear Tess, I read your thread about side-effects. You must be looking forward to the chemotherapy coming to an end. I know I was demob happy by my last session and couldn't wait for it to finish.

Waiting for results is beastly. I always tell myself that the dye is cast so there's no point in worrying about it ... and I try to think of some treats that will take my mind off it.

Hi Annie,

I am looking forward to it coming to an end, but a bit anxious about what comes next! Have to wait and see what the scan says, but will probably have to go straight back onto another regime but am hoping for a couple of weeks or so off (fingers crossed)
