Trovagene’s investigational oral therapy onvansertib helps overcome resistance to Zytiga (abiraterone acetate) in men with metastatic castration-resistant prostate cancer (mCRPC), latest data from a Phase 2 clinical trial show. These results also highlighted that adding onvansertib to a standard Zytiga (by Janssen Biotech) and prednisone combination led to treatment responses in patients carrying markers associated with resistance to Zytiga and other androgen receptor signaling (ARS) inhibitors. “Observing efficacy in patients that have tumors exhibiting known mechanisms of resistance to ARS inhibitor, Zytiga, suggests that onvansertib’s activity could extend to overcoming resistance to other ARS inhibitors, such as Xtandi and Erleada,” Mark Erlander, MD, Trovagene’s chief scientific officer, said in a press release.

Dana Farber in Boston has asked my husband if he would be interested in participating in this trial. Last week, he had blood work done, a EKG and two scans and now we are waiting to hear back from them about begining this new drug. I hope I will be able to report good news in the future!

Good for him.

I just looked at the study and my husband is ineligible because he was already on Xtandi. Also, he has gone from using prednisone to dexamethasone with his abiraterone.

Switching from prednisone to dexamethasone disqualifies? I wonder if this also means it may not work if you've already switched steroids, as my husband has too.

The Doctor's at Dana Faber also said my husband must stop taking the dexamethasone immediately and switch back to prednisone before beginning the trial. We got the call yesterday that he has been accepted, and we will be starting the trial on March 5.

Good for you. Are living in Boston or have to travel and if so how long the stay? Good luck. I think I might want to contact them.

We live in Southern New Hampshire. It's about a 1 hour drive to Dana Faber. First appointment is at 1:30 and the last is at 5:15, so it only involves day trips. I believe we will have to go to Boston every other week for only a few hours.

Waiting to hear if he has to wait 4 weeks before starting the trial because he was taking grape seed extract as a supplement!

We got the results today from taking 2 rounds of Onvansertib. It appears to be working. His PSA has dropped by 30 points! He has had no side effects from the drug. He adds 4 pills a day (based on your height and weight) to his Zytiga for 5 days, and then takes only Zytiga for the next 14 days. So far, so good!

Wonderful news!! Did your husband test for any markers that would indicate he's a good candidate for the ovansertib trial? Also, are you seeing this effect his blood counts, particularly his platelets?

He did have genetic testing done. I know he did not have the BRCA2 mutation. I'm not exactly sure why they thought he was a good candidate except for the fact that there is no cancer in any of his organs except for the bones. All his bloodwork has not been posted on line yet, so I don't know what his platelet numbers are. We just got a call from the hospital this afternoon saying things looked good and his PSA is dropping. I'll keep you posted as we get more information.

His platlets increased from 148 to 190.

That's great! I read Ovansertib can lower blood counts, but if it's working well for your husband then it makes sense that the blood counts could also improve. I hope this keeps working for a long time!
